The Super Rugby Trans-Tasman five-week tournament will start on 14 May 2021 in Dunedin, where Highlanders will play against QLD Reds at Forsyth Barr Stadium.
Crusaders, Highlanders Hurricanes, Blues, and Chiefs will play in the first round at their home ground against New Zealand's five super rugby clubs, Reds, NSW Waratahs, Brumbies, Melbourne Rebels, and Western Force.

On 19 June 2021, the final match will take place between the two highest-ranked teams at the combined table.
The schedule 2021 includes two family-friendly matches in New Zealand, which will start at 4:35 P.M. The Blues will battle with the Waratahs at Eden Park in Round 2, and the Rebels will compete with the Chiefs in round 4 at FMG Stadium Waikato.
As already declared, the third round will be a super round, and all matches will be played in one location. NZR and Australian Rugby will declare the venues and other details of the Super Round in due course.
